This glossary was made for the purpose of a better understanding of fifteen words commonly used in Bacteriology. These are the words biotechnologists, such as myself, employ throughout a day to day basis at a laboratory.

archaeon
noun
An archaeon is a single-celled, prokaryotic microorganism belonging to the domain Archaea, constituting a distinct "third form of life" separate from Bacteria and Eukarya.
Example: The archaeon survived extreme salinity thanks to specialized membrane lipids found only in Archaea.

bunsen burner
noun
A Bunsen burner is a common piece of laboratory equipment that produces a single, open gas flame for heating, sterilization, and combustion.
Example: A BUNSEN BURNER creates and aseptic environment by creating a column of hit rising air.

carboxysome

Carboxysomes are specialized bacterial microcompartments (BMCs) found in cyanobacteria and many autotrophic bacteria that function as protein-based organelles to enhance carbon fixation.
Example: Cyanobacteria rely on the carboxysome to concentrate carbon dioxide and improve the efficiency of carbon fixation.

A division of microorganisms that are related to the bacteria but are capable of photosynthesis. They are prokaryotic and represent the earliest known form of life on the earth.
Example: The presence of DNA in chloroplasts is explained by the endosymbiosis theory as originating from primitive CYANOBACTERIA during endocytosis.

endospore cortex

The endospore cortex is a thick, specialized layer of peptidoglycan located between the inner and outer membranes of a bacterial endospore.
Example: The endospore cortex provides structural protection that allows dormant spores to survive extreme environmental stress.

fimbria
noun
Fimbriae are short, hair-like protein appendages found on the surface of many bacteria, particularly Gram-negative bacteria, used primarily for adhesion to host cells, surfaces, and other bacteria.
Example: Each fimbria on the bacterial surface helps the cell adhere tightly to host tissues during colonization.

glycocalyx

The glycocalyx is a delicate, sugary, gel-like layer coating the surface of animal cells, bacteria, and epithelial cells.
Example: The thick glycocalyx surrounding the bacterium protects it from desiccation and enhances its ability to form biofilms.

gram stain

Gram staining is a rapid laboratory technique used to classify bacteria into two broad groups—Gram-positive (purple) and Gram-negative (pink/red)—based on their cell wall composition.
Example: The most common differential stains are: GRAM STAIN Acid fast stain Spore stain Capsule Stain The GRAM STAIN The structural differences between Gram + and Gram - cells. From The GRAM STAIN is usually the first test performed for the identification of bacteria.

Example: The magnetosome allows magnetotactic bacteria to align with Earth’s magnetic field and navigate toward optimal oxygen levels.

nucleoid occlusion

Nucleoid occlusion is a bacterial safety mechanism that prevents the cell’s division machinery (the Z-ring) from forming near the chromosome (nucleoid), ensuring the cell divides at the mid-cell rather than cutting through the DNA.
Example: Nucleoid occlusion prevents the division septum from forming over the bacterial chromosome, ensuring proper cell division.

operon

An operon is a functional unit of genomic DNA in prokaryotes (bacteria and archaea) containing a cluster of genes under the control of a single promoter.
Example: These evidence include similarities in patterns of gene organisation, which is the OPERON arrangement, gene expression, which consists of promoter, RNA polymerase and ribosome structure; nature and function of protein synthesis factors; use of N-formylmethionyl-tRNA as initiator in translation; antibiotic sensitivity in transcription and translation (which will be explored further) and gene structure which comprises of rRNA, tRNA and protein 4 .

A shallow, circular, transparent dish with a flat lid, used for the culture of microorganisms.
Example: Place a filter paper pad in a sterile PETRI DISH and pour 2-3 ml of nutrient medium over the pad.

protoplast
noun
The protoplasm of a living plant or bacterial cell whose cell wall has been removed.
Example: After enzymatic digestion of the cell wall, the bacterium became a protoplast that could be used for DNA uptake experiments.

spheroplast
noun
A spheroplast is a bacterial or yeast cell with its cell wall partially removed, typically via enzymatic action (e.g., lysozyme) or antibiotics, causing it to adopt a characteristic spherical shape.
Example: The antibiotic treatment weakened the bacterial cell wall, producing a fragile spheroplast that was highly sensitive to osmotic pressure.
